SRX7138444: target enrichment of UCEs and exons from anthozoans

Design: We prepared DNA libraries using the Kapa Biosystems Hyper Prep Kit with dual indexed iTru adapters (Glenn et al. 2016). The resulting DNA libraries were combined in equimolar pools of 5 to 8 samples. Pooled libraries were enriched for UCE loci using the anthozoa-v1 bait set (Quattrini et al. 2017) synthesized by MYcroarray (Ann Arbor, MI). We quantified each enriched pool, post- PCR amplification, using a Qubit fluorometer and checked them for adapter contamination using a Bioanalyzer. If adapters were found, a 1X bead cleanup was performed. After cleaning up any remaining adapter contamination, we qPCR quantified each pooled library using the Kapa qPCR quantification kit, and we combined pooled libraries prior to sequencing.
